Pain is a noxious sensation caused by tissue damage, which can severely interfere with a person’s quality of life. Although numerous analgesics are available for eradicating pain, there remain limitations in terms of safety and efficacy. This review focuses on arachidonoyl-serotonin (AA-5-HT) − an endogenous lipid with a putative antinociceptive effect. After detailed investigation, previous studies have revealed that AA-5-HT can stimulate the cannabinoid system, which results in the inhibition of pain sensation. Moreover, AA-5-HT can inhibit the action of TRPV1, which is a nonselective cation channel that mediates pain signals in the nervous system. This dual effect makes AA-5-HT a potentially safe and potent analgesic. This review summarizes the roles of the cannabinoid system and TRPV1 in pain sensation, and the function of AA-5-HT in pain modulation.

Liposomes are spherical vesicles composed of a bilayer (uni-lamellar) and/or a concentric series of multiple bilayer (multi-lamellar) separated by aqueous compartments formed by amphipathic molecules, such as phospholipids. Liposomes deliver drugs via encapsulation in inner aqueous or lipid compartments and have advantages such as control of drug release and distribution, alleviation of toxicity, and drug targeting. Since the launch of the first liposomal drug Doxil??, a variety of products using a liposomal drug delivery system have been developed and commercialized. Recently, several drug candidates developed with sophisticated and diverse technologies have been undergoing clinical trials. In this review, the pharmaceutical characteristics of approved liposomal drugs are overviewed, and the recent advances in liposome technology through candidates in clinical trials are highlighted.

BIOVITA 3 bacterial species (Clostridium butyricum, Lactobacillus sporogenes, and Bacillus subtilis) are spore forming probiotic bacteria that are blended as granules. Unlike lactic acid bacteria, which account for most of the available probiotics, the BIOVITA 3 bacterial species are easy to store and stably transit to the intestine, where they promote intestinal health owing to the unique features of the individual strains. The BIOVITA 3 bacterial species differ in their proliferation in different regions of the intestinal tract because of their differing oxygen demands. The aerobic Bacillus subtilis mainly proliferates in the upper small intestine, which has a high oxygen concentration. The facultative anaerobe Lactobacillus sporogenes proliferates well in the small intestine. The anaerobic Clostridium butyricum grows best in the large intestine, which has a relatively low oxygen concentration. In addition, each bacterial species displays positive effects on promoting the growth of bacteria resident in the lower intestine. The differential proliferation of the BIOVITA 3 bacterial species influences intestinal regulation throughout the intestine from the small to the large intestine. The species also improve intestinal regulation through mutual interactions that increase the growth of other beneficial bacteria.

Omega fatty acids play an important role as biomarkers of chronic inflammatory diseases such as asthma, chronic obstructive pulmonary disease, rheumatoid arthritis, and inflammatory bowel disease. The purpose of our study was to develop a new analytical method for the detection of omega fatty acids from dried serum spots. We developed an ultrahigh performance liquid chromatography-electrospray ionization tandem mass spectrometry (UHPLC-ESI-MS/MS)-based method in the multiple reaction monitoring (MRM) mode with negative ionization. The detected MRM transitions for five omega fatty acids are as follow: α-linolenic acid (m/z=277.20→277.20), eicosapentaenoic acid (m/z=301.10→257.30), docosahexaenoic acid (m/z=327.00→283.15), arachidonic acid (m/z=303.20→259.25), and docosapentaenoic acid (m/z= 329.10→285.25). The calibration curve showed an excellent linearity within 0.1-2.5 μg/mL range (R2=0.9947~0.9994). The analytical methods howed excellent sensitivity (LOD: 0.005-0.01 μg/mL, LOQ: 0.03-0.1 μg/mL), which would allow for the targeted analyses of omega fatty acids. The recoveries of the omega fatty acid from the dried serum spots were 92.6%- 110.4% (RSD: ±0.2%-9.6%) and the retention time was within 5 min. This improved method offers rapid and sensitive quantification of omega fatty acids on dried serum spots using a small volume of serum and is expected to be applied to various biological specimens.

Proton Pump Inhibitors (PPIs) are drugs that inhibit gastric acid secretion and are widely used globally. With the aging population and the rising prevalence of gastrointestinal diseases in Korea, the number of patients using PPIs is increasing. Therefore, we investigated the current state of PPI utilization at the national level using the nationwide database, Healthcare Bigdata Hub compiled by the Health Insurance Review and Assessment Service (HIRA). We extracted the statistics from 2016 to 2020 and re-organized them into monthly utilization by treatments. The cost and volume of use by region, provider type, and diagnostic code were extracted for five PPIs: pantoprazole, rabeprazole, omeprazole, esomeprazole, and lansoprazole. The volume of use was converted into Defined Daily Dose (DDD), which is the standardized unit of utilization. Esomeprazole and rabeprazole were the most commonly used PPIs, and esomeprazole has shown a sharp increase recently. The crude utilization of each region was the highest in Seoul, but that in Daegu and Jeonbuk was the highest after adjusting for the population size. The utilization of PPIs has increased in all diagnostic codes, and Gastroesophageal Reflux Disease (GERD) accounts for the highest proportion. Despite emerging concerns on the adverse outcomes of long-term PPI utilization, their consumption has increased in recent years. These results on the variations by region and trends by diagnostic code thus address a good basis for future research and policies regarding PPIs.

This study aimed to examine the protective effect of the water extract from Spatholobi Caulis (SC) on rats with acute liver injury (ALI) induced by thioacetamide (TAA) and its underlying mechanism. ALI was induced by intraperitoneal injection with TAA for 3 days in a row and the rats were administered SC (100 or 200 mg/kg) 1 h 30 min prior to the TAA treatment. Myeloperoxidase (MPO) was measured in serum and malondialdehyde (MDA) was measured in serum and liver tissue, as markers related to oxidative stress. Further, inflammation-related proteins and apoptosis proteins were investigated using western blot analysis. SC treatment significantly reduced the levels of MPO and MDA. Further, SC treatment significantly reduces the mitogen-activated protein kinase (MAPK) signaling pathway, including p-JNK, p-ERK, and p-p38, and reduces the expression of pro-inflammatory signaling factors. Furthermore, it effectively decreased the expression of Bax, Cytochrome C, and Cleaved Caspase-3 in liver tissue. Taken together, SC treatment alleviate0d the severity of ALI via inhibition of inflammation through both the MAPK pathway and Matrix metalloproteinase (MMPs)-dependent apoptosis.

Despite the high efficacy and ease of use of contraception, the rate of oral contraceptive use in the Republic of Korea remains low. This study aimed to investigate Korean women’s knowledge level and social perception toward oral contraceptives as well as their perspectives on pharmacy visits. This cross-sectional study was conducted as a 37-item online survey questionnaire on Korean women between the ages of 20 and 49. A total of 1,076 women participated in the study, and 52.0% of the participants have experienced taking oral contraceptives (“ever users”). Among oral contraceptive users, the most common reasons for taking oral contraceptives were menstrual cycle delay (67.6%) and contraception (44.7%). Regarding the social perception on oral contraceptives, single women felt more social prejudice and felt less comfortable talking about oral contraceptives compared with married women, and similar pattern was observed in younger women compared to older women. Oral contraceptive users had higher knowledge levels compared never users, but the knowledge of adverse events appeared to be low in all respondents. Ever users felt more comfortable with the pharmacy visit to purchase oral contraceptives, whereas never users felt more self-conscious of their surroundings. This study suggests that pharmacists play a significant role in oral contraceptive counseling for women, which should be conducted in a comfortable and private setting to ensure safe and effective use of oral contraceptives.

The purpose of this study was to investigate and analyze the application status and system introduction perceptions of quality-by-design (QbD) in domestic pharmaceutical companies and confirm the necessity of introducing an effective QbD system. Currently, QbD-applied drug development is an international trend, and the introduction of QbD domestically is urgent because domestic pharmaceutical companies need QbD data even when exporting overseas. Therefore, 51 domestic pharmaceutical companies were surveyed to analyze their perceptions and opinions on the current status of QbD application and the introduction of the system. The results showed that most pharmaceutical companies, except 4.7% of total respondents, recognized the need for QbD for the following reasons: to improve the quality of medicines, responding to requests from overseas regulatory authorities when exporting products, problem-solving drug development and production, and proactively responding to the introduction of the QbD system by the Ministry of Food and Drug Safety. Although they are aware of its need, respondents are not implementing QbD due to cost and time issues and state that it is challenging to apply the QbD example in practice due to the method and amount of experimentation. Thus, the results of this study revealed that domestic pharmaceutical companies need QbD experimental and process guidelines and practical QbD examples. These results can be used as the foundation for the introduction and stable settlement of a domestic QbD system in the future.

Melanoma is a belligerent cutaneous malignancy. Multiple therapeutic options, including immunotherapy and targeted therapy, have shown remarkable promise in treating metastatic melanoma. Exosomes containing programmed cell death ligand 1 (PD-L1) secreted from melanoma bind to programmed cell death 1 (PD-1) on the surface of CD8+ T cells. As a result, melanoma not only evades the immune surveillance of CD8+ T cells but also inhibits their activation and facilitates the growth and proliferation of cancer cells. In this study, we identified nicardipine, an FDA-approved L-type calcium channel inhibitor, which downregulates the expression of proteins involved in exosome biogenesis and secretion as well as exosomal PD-L1 secretion, from metastatic melanoma cells. Furthermore, nicardipine suppresses PD-L1 expression after interferon-γ-mediated stimulation and enhances CD8+ cell function, similar to the function of PD-L1- blocking antibodies in cancer therapies. This could lead to the development of next-generation anticancer drugs for the treatment of metastatic melanoma.

As the market of the functional foods is expanding to general foods, more efficient and accessible methods are required to provide quantitative and scientific evidences. In this study, we conceptualized and established a functional reporting model with logistic regression using the clinical data retrieved from the published literature. The model was validated with two active ingredients, vitamin E and lycopene. A search through PubMed using “tocopherol” or “tocotrienol” or “Vitamin E” as the keywords resulted in 45 data points corresponding to 35 studies in which the antiinflammatory efficacy of vitamin E was evaluated. The other search through PubMed using “lycopene” as a keyword resulted in 39 data points corresponding to 29 studies in which the antioxidant efficacy of lycopene was evaluated. We used C-reactive protein (CRP) or high sensitivity C-reactive protein (hs-CRP) as the inflammatory markers, and DNA damage, lipid, and protein oxidation for the measurement of antioxidant efficacy. According to our model, daily intake of at least 584.05 mg of vitamin E and 17.85 mg of lycopene is recommended to induce the functional effects. In conclusion, we anticipate that this approach to the functional report modeling would greatly expand the market of functional foods, especially for the native crops.

3,6-Disubstituted pyridazine analogs (2a and 2b) were synthesized from commercially available 3,6- dichloropyridazine in two steps. The synthesized compounds were evaluated for their GPR119 agonistic activity. Both compounds exhibited much stronger EC50 values than that of oleylethanolamide (OEA) and were proved to be partial agonists. These results indicate that the pyridazine ring can be used as a potential heterocycle scaffold for GPR119 agonists.
